Is it necessary to fill the engine block with coolant before first start up or is filling the radiator and overflow tank sufficient

No, you need to fill the block too so it's not dry, then burp the block as best as possible. I'd suggest filling the block through the thermostat outlet on the intake manifold. It might take a little time, but this will make sure the block isn't empty when you start the motor, and risk a severe overheat because of air trapped in the block.
